The crystal structure of the Fab fragment of the antibody in complex with human IL-34 was determined. The original antibody was reformatted as an IgG molecules and the binding affinity of both formats measured by BLI experiments (Kd= 17 nM and 120 pM respectively). The antibody blocked the biological activity of IL-34 in human monocyte viability. The antibody neutralized IL-34 in a proliferation assays (Ma et al., 2012; PMID: 22483114).
