Precipitation assays. In immunoelectrophoresis use 2 microl human serum against 120 microl antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 microl antiserum in a 3 mm diameter center well and 10 microl of the antiserum in 3 mm diameter peripheral wells. In single radial immunodiffusion (Mancini) use 1% antiserum in the gel. Preciptin titre not less than 1:8 when tested against normal human serum in agar block immunodiffusion titration.
