    Immunohistochemistry: LS-B8215 in an IHC-plus antibody, validated by our pathologists for use in IHC against formalin-fixed, paraffin-embedded human tissues (IHC validation procedure). A concentration of 5 ug/ml is recommended. ELISA: To detect hEndostatin by sandwich ELISA (using 100 ul/well antibody solution) a concentration of 0.5-2 ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human Endostatin (LS-C104550) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant hEndostatin. Western Blot: To detect hEndostatin by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hEndostatin is 1.5-3 ng/lane, under either reducing or non-reducing conditions.. Immunofluorescence staining of A549 cells with CSB-PA005725LA01HU at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. Figure 1. Western blot analysis of COL18A1 using anti-COL18A1 antibody (A01302-1). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human COLO-320 whole cell lysates, Lane 2: human placenta tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-COL18A1 antigen affinity purified polyclonal antibody (Catalog # A01302-1) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for COL18A1 at approximately 180KD, 20KD. The expected band size for COL18A1 is at 178KD.
