
WB (peptide competition) analysis of CHO-T cells transiently transfected with wild-type human IRS-1 and treated with 100 ng/mL TPA for 30 minutes using GTX24865 IRS1 (phospho Ser312) antibody prior incubated with no peptide (Lane 1~3) , the non-phosphopeptide corresponding to the phosphopeptide immunogen (Lane 4), a generic phosphoserine-containing peptide (Lane 5), or the phosphopeptide immunogen (Lane 6) . The data show that only the immunogen phosphopeptide blocks the signal, demonstrating the specificity of the antibody. The membrane treated with phosphatase (Lane 1) eliminates the signal further verifying that the antibody is phospho-specific.
