WB (peptide competition) analysis of NIH3T3 cells treated with 50ng/mL PDGF for 15 minutes (Lane 2-5) using GTX25615 MEK1 (phospho Thr386) antibody with the non-phosphopeptide corresponding to the phosphopeptide immunogen (Lane 3), a generic phosphothreonine-containing peptide (Lane 4), or the phosphopeptide immunogen (Lane 5) control. The data show that only the immunogen phosphopeptide blocks the signal, demonstrating the specificity of the antibody.

    The protein encoded by this gene is a member of the dual specificity protein kinase family, which acts as a mitogen-activated protein (MAP) kinase kinase. MAP kinases, also known as extracellular signal-regulated kinases (ERKs), act as an integration point for multiple biochemical signals. This protein kinase lies upstream of MAP kinases and stimulates the enzymatic activity of MAP kinases upon wide variety of extra- and intracellular signals. As an essential component of MAP kinase signal transduction pathway, this kinase is involved in many cellular processes such as proliferation, differentiation, transcription regulation and development. Figure 1. Western blot analysis of MEK1/MAP2K1 using anti-MEK1/MAP2K1 antibody (A00292). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human Hela whole cell lysates, Lane 2: human Daudi whole cell lysates, Lane 3: rat lung tissue lysates, Lane 4: rat brain tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-MEK1/MAP2K1 antigen affinity purified polyclonal antibody (Catalog # A00292) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for MEK1/MAP2K1 at approximately 45 kDa. The expected band size for MEK1/MAP2K1 is at 45 kDa.
