  • Scientific Description
    The protein encoded by this gene is involved in both the synthesis and degradation of fructose-2,6-bisphosphate, a regulatory molecule that controls glycolysis in eukaryotes. The encoded protein has a 6-phosphofructo-2-kinase activity that catalyzes the synthesis of fructose-2,6-bisphosphate, and a fructose-2,6-biphosphatase activity that catalyzes the degradation of fructose-2,6-bisphosphate. This protein regulates fructose-2,6-bisphosphate levels in the heart, while a related enzyme encoded by a different gene regulates fructose-2,6-bisphosphate levels in the liver and muscle. This enzyme functions as a homodimer. Two transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Figure 1. Western blot analysis of PFKFB2 using anti-PFKFB2 antibody (A05977). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30ug of sample under reducing conditions. Lane 1: human K562 whole cell lysates, Lane 2: human HepG2 whole cell lysates, Lane 3: human Caco-2 whole cell lysates, Lane 4: human Hela whole cell lysates, Lane 5: human A431 whole cell lysates, Lane 6: human PC-3 whole cell lysates, Lane 7: rat heart tissue lysates, Lane 8: rat PC-12 whole cell lysates, Lane 9: mouse heart tissue lysates, Lane 10: mouse RAW264.7 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PFKFB2 antigen affinity purified polyclonal antibody (Catalog # A05977) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PFKFB2 at approximately 58KD. The expected band size for PFKFB2 is at 58KD.
