AXL1717 (picropodophyllin, Picropodophyllotoxin, PPP) is a non-competitive, potent, and specific inhibitor of IGF-1Ralfa/bèta with IC50 of 1 nM. Picropodophyllin binds to the intracellular tyrosine kinase domain of IGF-1R, inducing a conformational shift to an inactive state, thereby effectively inhibiting its autophosphorylation and downstream signal transduction.
