    FLIP is an apoptosis regulator protein which functions as a crucial link between cell survival and cell death pathways in mammalian cells and acts as an inhibitor of TNFRSF6 mediated apoptosis. A proteolytic fragment (p43) is likely retained in the death-inducing signaling complex (DISC) thereby blocking further recruitment and processing of caspase-8 at the complex. Full length and shorter isoforms have been shown either to induce apoptosis or to reduce TNFRSF-triggered apoptosis. FLIP lacks enzymatic (caspase) activity. FLIP is highly expressed in skeletal muscle, pancreas, heart, kidney, placenta and peripheral blood leukocytes. - Monoclonal Antibody. Mouse NLRP1b (mouse) (full-length and UFA-CARD fragment) is detected in Raw264.7 cells using NLRP1b, mAb (2A12) (Prod. No. AG-20B-0084). Method: Cell extracts (about 30microg) from the mouse cells Raw264.7 (lane 1) or Raw264.7 NLRP1b KO (lane 2) are separated by SDS-PAGE under reducing conditions, transferred to nitrocellulose and incubated with the NLRP1b, mAb (2A12) (1microg/ml). After incubation with anti-mouse Alexa 633 conjugated antibody, proteins are visualized by a fluorescence detection system at 700nm. Figure 1: Immunoblot analysis of beta-tubulin protein glutamylation using anti-beta-Tubulin, mAb (AXO45) (Prod. No. AG-20B-0085). Method: HEK-293T cells are grown in standard culture conditions, transfected with plasmids expressing the different glutamylases, TTLL4 and TTLL6 and are run on a 10% SDS-PAGE (specific protocol to separate alpha- and beta-tubulin (Magiera & Janke, 2013)). The proteins are transferred to a nitrocellulose membrane and detected by standard immunoblot protocol using the AXO45 (1:1000) in PBS containing 0.1% Tween-20 for washing steps and 2.5% fat free milk for antibody incubation. In all the cell extracts, AXO45 specifically detects beta-tubulin, with no cross-reactivity to alpha-tubulin. Glutamylation added by either TTLL4 or TTLL6 appears to not affect this detection.
