    GAPDH (Glyceraldehyde 3-phosphate dehydrogenase) is an enzyme of ~37kDa which is a tetramer composed of four identical 37-kDa subunits, containing a single catalytic thiol group each and critical to the enzymes catalytic function. GADPH catalyzes the sixth step of glycolysis and thus serves to break down glucose for energy and carbon molecules. In addition to this long established metabolic function, GAPDH has recently been implicated in several non-metabolic processes, including transcription activation, initiation of apoptosis, ER to Golgi vesicle shuttling, DNA repair. membrane fusion, microtubule bundling and age-related actions in neurodegenerative diseases. - Recombinant Antibody. This antibody reacts to GAPDH. RM114 shows strong reactivity to Human/Monkey GAPDH. RM114 shows weaker reactivity to Mouse, Goat GAPDH and may require higher antibody dilution usage. Applications: WB, IP, ICC, IHC, FACS, ELISA . Source: Rabbit. Liquid. 50% Glycerol/PBS with 1% BSA and 0.09% sodium azide. Figure 1. Western blot analysis of GAPDH using anti-GAPDH antibody (A00227-1). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human Hela whole cell lysates, Lane 2: human CACO-2 whole cell lysates, Lane 3: human CCRF-CEM whole cell lysates, Lane 4: rat brain tissue lysates, Lane 5: rat liver tissue lysates, Lane 6: mouse brain tissue lysates, Lane 7: mouse liver tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GAPDH antigen affinity purified polyclonal antibody (Catalog # A00227-1) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for GAPDH at approximately 36 kDa. The expected band size for GAPDH is at 36 kDa.
