- Application Supplier NoteThe original antibody detected CK 8 by western blot from BT20 breast cancer cells. Immunofluorescence was performed on BT20 cells using this antibody, showing the carboxyl terminus of CK 8 was exposed on the surfaces of breast cancer cells. The Fab fragment of the antibody was used for inhibition experiments on breast cancer cell lines BT20, MDA-MB-157, MCF-7 and purified CK 8. Results showed a decreased plasminogen binding in all the experiments. The Fab fragment was used to study the role of cell-surface CK 8 in promoting the activation of plasminogen by t-PA in MCF-7 cells; preincubation of MCF-7 cells with the Fab fragment caused a concentration-dependent decrease in the ability of the cell cultures to promote plasminogen activation (Hembrough et al, 1996; pmid:8810346).
