The binding reactivity of this antibody for mouse sclerostin was tested using ELISA. This antibody binds mouse and human sclerostin with a binding affinity of Kd= 46nM and Kd=260 nM respectively. This antibody efficiently neutralizes sclerostins Wnt inhibitory activity. In vitro interaction analysis using sclerostin variants revealed that this neutralizing Fab binds to sclerostins flexible second loop, which has been shown to harbor the LRP5/6 binding motif (PMID: 27558933).
