Unboiled various whole cell extracts (30 μg) were separated by 12% SDS-PAGE, and the membrane was blotted with Claudin 5 antibody [HL3156] (GTX640666) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody (GTX213110-01) was used to detect the primary antibody. Corresponding RNA expression data for the same cell lines are based on Human Protein Atlas program.

Figure 1. IHC analysis of Claudin 5/CLDN5 using anti-Claudin 5/CLDN5 antibody (A03260-3). Claudin 5/CLDN5 was detected in paraffin-embedded section of human rectal cancer tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2microg/ml rabbit anti-Claudin 5/CLDN5 Antibody (A03260-3) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) (Catalog # SA1022) with DAB as the chromogen.
