Figure 1. Western blot analysis of Glycogen synthase 1/GYS1 using anti-Glycogen synthase 1/GYS1 antibody (A03512-3). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: rat skeletal muscle tissue lysates, Lane 2: mouse skeletal muscle tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Glycogen synthase 1/GYS1 antigen affinity purified polyclonal antibody (Catalog # A03512-3) at 0.25 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Glycogen synthase 1/GYS1 at approximately 84-90 kDa. The expected band size for Glycogen synthase 1/GYS1 is at 84 kDa.

Affinity Purified Phospho-specific antibody to human muscle Glycogen Synthase at pS640 (GTX22479) was used at a 1:1000 dilution to detect human muscle GS by Western blot. Approximately 12 ul of a mouse cardiac myocyte lysate was loaded per lane on a 4-20% Criterion gel for SDS-PAGE. Samples were either mock treated (lanes 1 and 5) or insulin treated at 10 nM, 100 nM and 1 microM (lanes 2, 3 and 4 respectively) for 15 or CLA treated at 4nM, 20 nM or 100 nM (lanes 6,7 and 8 respectively) for 45. After washing, a 1:5,000 dilution of HRP conjugated Gt-a-Rabbit IgG (GTX27090) preceded color development using Amershams substrate system.
