    Peptide ELISA: antibody detection limit dilution 1:16000. Western blot: Preliminary experiments gave an approx 75Da band in Human Heart lysates after 1 ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 58.5kDa according to NP_006203.2 and 54.4kDa according to NP_001018063.1. The 75kDa band was successfully blocked by incubation with the immunizing peptide.. IHC, IHC-P (2.5 - 3.75 µg/ml), Peptide-ELISA (1:16000) Peptide ELISA: antibody detection limit dilution 1:16000. Figure 1. Western blot analysis of PFKFB2 using anti-PFKFB2 antibody (A05977). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30ug of sample under reducing conditions. Lane 1: human K562 whole cell lysates, Lane 2: human HepG2 whole cell lysates, Lane 3: human Caco-2 whole cell lysates, Lane 4: human Hela whole cell lysates, Lane 5: human A431 whole cell lysates, Lane 6: human PC-3 whole cell lysates, Lane 7: rat heart tissue lysates, Lane 8: rat PC-12 whole cell lysates, Lane 9: mouse heart tissue lysates, Lane 10: mouse RAW264.7 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PFKFB2 antigen affinity purified polyclonal antibody (Catalog # A05977) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for PFKFB2 at approximately 58KD. The expected band size for PFKFB2 is at 58KD.
