    Rabbit polyclonal to TET1.TET1 is a methylcytosine dioxygenase that catalyzes the conversion of the modified genomic base 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC). Methylation at the C5 position ofcytosine bases is an epigenetic modification of the mammalian genome which plays an important rolein transcriptional regulation. TET1 might initiate a process leading to cytosine demethylation throughdeamination into 5-hydroxymethyluracil (5hmU) and subsequent replacement by unmethylatedcytosine by the base excision repair system. TET1 preferentially binds to CpG-rich sequences atpromoters of both transcriptionally active and polycomb-repressed genes. By controlling the levels of5mC and 5hmC at gene promoters, it may regulate the gene silencing induced by cytosinemethylation. It has been shown that TET1 plays an important role in embryonic stem cellsmaintenance and regulating the balance between pluripotency and lineage commient.

Figure 1. Western blot analysis of TET1 using anti-TET1 antibody (A00603-2). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human HEK293 whole cell lysates, Lane 2: human K562 whole cell lysates, Lane 3: human SW620 whole cell lysates, Lane 4: human HepG2 whole cell lysates, Lane 5: monkey Cos-7 whole cell lysates, Lane 6: human Raji whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TET1 antigen affinity purified polyclonal antibody (Catalog # A00603-2) at 0.25 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TET1 at approximately 235KD. The expected band size for TET1 is at 235KD.
