The scFv fragment of the antibody was able to bind the recombinant human Agr2 protein in ELISA analysis. The binding affinity of the scFv fragment to human Agr2 was measured by surface plasmon resonance (Kd= 4.6x10^-7 M). Immunofluorescence analysis of colon biopsy samples from a healthy donor was performed to verify the ability of CPR02 to recognize the human Agr2 antigen. Results showed the antibody revealed significant staining surrounding the crypts of the intestinal tissue. Further, the antibody was used for immunofluorescent staining of freshly frozen human biopsy samples of Ulcerative Colitis, the antibody produced a strong intensity signal in the colonic crypts region. The antibody was able to recognise the Agr2 antigen in vivo, as demonstrated by the results of the autoradiography of the complete intestinal tract of a mouse having colitis, injected with radiolabelled antibody (US20190309060A1).
