    This antibody recognizes and binds the Notch negative regulatory region (NRR) of Notch 1.

    This antibody is capable of immunoprecipitating Notch 1 but not Notch 4. The binding affinity of this antibody towards Notch1 was checked using ELISA. The binding of this antibody to full length Notch 1 overexpressed on HEK cells was confirm using flow cytometry. Flow cytometry was also used to study the expression of Notch 1 on cancer cell lines like LS-1034, TALL-1, BxPC3, and Colo_205. This antibody does not compete with DLL4 for binding site on Notch1. The original human IgG1 version of this antibody is capable of inhibiting Jag2 mediated Notch1 signalling with an EC50 value of 0.13+/-0.09 nM. This antibody partially inhibited Hes1 transactivation. WC75 also inhibited the proliferation of NOTCH1 mutated T-ALL cell lines such as DND41 and KOPT-K1 (PMID: 20161710).
