GTX65635-pro CD40L protein (multimeric) does not need an enhancer to induce B cells activation. Method: PBL cells were incubated in 96-well plates (2x10 5 cells/well in 100μl RPMI supplemented with 10% FCS) for 24 hours at 37oC with the indicated concentration of CD40L (multimeric) or CD40L in the presence and absence of 1μg/ml Enhancer. Cells were washed with PBS and stained with CD86-PE antibody and CD19-FITC antibody, and then analyzed by flow cytometry.

Deglycosylation of GTX65638-pro rat progranulin (DDDDK tagged) protein. To examine the deglycosylation of rat Progranulin (DDDDDK tagged), 1 μg of rat progranulin is denatured with 1X glycoprotein denaturing buffer at 100oC for 10 minutes. After the addition of NP-40 and G7 reaction buffer, twofold dilutions of PNGase F are added and the reaction mix is incubated for 1 or 3 hours at 37oC. Separation of reaction products is visualized by immunoblotting using anti-DDDDDK-HRP antibody.
