  • Scientific Description
    This gene encodes a protein with a kinase-like domain and four ankyrin-like repeats. The encoded protein associates at the cell membrane with the cytoplasmic domain of beta integrins, where it regulates integrin-mediated signal transduction. Activity of this protein is important in the epithelial to mesenchymal transition, and over-expression of this gene is implicated in tumor growth and metastasis. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2013]

Immunofluorescence staining of MCF-7 cells with CSB-PA00999A0Rb at 1:66, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).

Figure 1. Western blot analysis of Integrin linked ILK using anti-Integrin linked ILK antibody (A02932-2). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human MCF-7 whole cell lysates, Lane 2: human Hela whole cell lysates, Lane 3: human U-87MG whhole cell lysates, Lane 4: human A431 whole cell lysates, Lane 5: human U20S whhole cell lysates, Lane 6: human PC-3 whhole cell lysates, Lane 7: rat heart tissue lysates, Lane 8: rat kidney tissue lysates, Lane 9: rat skeletal muscle tissue lysates, Lane 10: rat PC-12 whhole cell lysates, Lane 11: mouse heart tissue lysates, Lane 12: mouse kidney tissue lysates, Lane 13: mouse skeletal muscle tissue lysates, Lane 14: mouse C2C12 whhole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Integrin linked ILK antigen affinity purified polyclonal antibody (Catalog # A02932-2) at 0.25 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Integrin linked ILK at approximately 51 kDa. The expected band size for Integrin linked ILK is at 51 kDa.
