Western blot using GeneTex's Protein A purified anti-SPANX-C antibody shows detection of a band at ~17 kDa corresponding to SPANX-C present in a nuclear extract from VWM105 cells (left panel, arrowhead). VWM105 cells are derived from a human melanoma and are positive for SPANX proteins. Lane 2 shows reactivity with a purified recombinant SPANX-C fusion protein. The right panel shows similar reactivity with purified recombinant SPANX-B, SPANX-C and SPANX-N proteins. Proteins were separated by SDS-PAGE, transferred to nitrocellulose, and probed with the primary antibody diluted to 1:1,000. IRDye?800 conjugated goat anti-Rabbit IgG [H&L] was used (left). IRDye is a trademark of LI-COR, Inc. Size estimation was made by comparison to prestained MW markers as indicated.

Western blot of SUMO-GFP fusion proteins cleaved by insect cell protein extracts. Anti-SUMO antibody, generated by immunization with recombinant yeast SUMO, was tested by western blot against several constructs of SUMO-GFP fusion proteins after cleavage by proteases in insect cell protein extracts. These constructs contained various linkers between the SUMO and GFP portion of the fusion proteins. Each sample was run twice. The left lanes each contain 2 μg E.coli expressed and purified SUMO-GFP fusion proteins after incubation with lysed cells (50 μg total protein) for 1 h. The right lanes contain the same fusion proteins incubated with the lysate in the presence of 2% SDS. After probing with anti-GFP antibodies the membranes were stripped of antibody using SDS-DTT solution for 30 m at 60o C and were then re-probed using the anti-SUMO antibody at a 1:1000 dilution incubated overnight at 4o C in 5% non-fat dry milk in TBST. Detection occurred using a 1:2000 dilution of HRP-labeled Donkey anti-Rabbit IgG.

Anti-Rub1 antibody, generated by immunization with full-length, recombinant yeast Rub1, was tested by western blot against a yeast cell lysate. A dilution of the antibody between 1:200 and 1:1,000 will show strong reactivity specifically with free Rub1 protein (indicated by arrow) and Rub1 conjugates. Western blot of ySUMO fusion protein. Anti-ySUMO antibody, generated by immunization with recombinant yeast SUMO, was tested by western blot against a SUMO-GFP fusion protein (lane 2). While the actual molecular weight of the fusion protein is 39 kDa, the protein migrates as a 49 kDa band (arrowhead). No reactivity is seen for lane 1 which contains His-tagged GFP protein. The membrane was blocked using BLOTTO. Primary antibody was used at a 1:1,000 dilution in BLOTTO. The membrane was washed and reacted with a 1:10,000 dilution of IRDyeR 800 Conjugated Affinity Purified Goat-anti-Mouset IgG (H&L).
