Mouse cortex lysate was immunoprecipitated with anti-TrkCT1 antibody and further blotted with affinity purified anti-TrkCT1. Lane 1 is wild-type cortex lysate, Lane 2 is Tamalin knock-out cortex lysate, and Lane 3 is TrkCT1 knock-out cortex lysate. The membrane was probed with the primary antibody at a 1:6,000 dilution.

Western blot using GeneTex's affinity purified anti-AHA1 antibody shows detection of AHA1 in Cos7 cells. For Lanes 2 and 4, Cos7 cells were transfected with pcDNA3-FLAG-AHA1. For Lanes 1 and 3, Cos7 cells were not transfected. Extracts (40 μg per lane) were electrophoresed and transferred to nitrocellulose. The membrane was probed with anti-AHA1 (lanes 1 and 2, 1:2,000 dilution) or anti-FLAG (lanes 3 and 4). The lower band seen in anti-AHA1 blotting (arrowhead) is endogenous AHA1.

Western blot using GeneTex's affinity purified anti-Cyclin E2 antibody shows specific detection of Cyclin E2. Cell extracts over-expressing mouse Cyclin E1 (lane 1) and Cyclin E2 (lane 2) were electrophoresed, transferred to nitrocellulose, and probed with the anti-Cyclin E2 antibody. The affinity purified antibody also detects endogenous Cyclin E2 in Skp2-/- MEF cells. (data not shown).

Western blot using GeneTex's affinity purified Anti-MDM2 (Rabbit) is shown to detect a band (arrow) corresponding to mouse MDM2 protein present in mouse MEF cells (lane 2) but not human kidney HEK293 cells (lane 1). Approximately 35 ug of lysate was separated by 4-20% Tris Glycine SDS-PAGE. After blocking the membrane with 5% normal goat serum, 0.5% BLOTTO in PBS, the membrane was probed for overnight at 4o with the primary antibody diluted to 1:500 in 1% normal goat serum, 0.1% BLOTTO in PBS. The membrane was washed and reacted with a 1:10,000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] for 45 min at room temperature (800 nm channel, green). Molecular weight estimation was made by comparison to prestained MW markers indicated at the right (700 nm channel, red). IRDye800 fluorescence image was captured using the OdysseyR Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Western blot using GeneTex's affinity purified anti-S.cerevisiae Mer2 antibody shows detection of phosphorylated and unphosphorylated Mer2 in wild type, phosphatase treated and mutant cells. Lane 1 contains Mer2-myc protein detected in wild type cells after first immunoprecipitating the protein using anti-myc antibody. Cells were harvested 4 h after the initiation of meiosis and therefore contain mostly phosphorylated Mer2. Lane 2 contains the same preparation after treatment with phosphatase. Lane 3 contains Mer2-S30A protein as a phosphorylation control. This antibody is reactive with both phosphorylated and unphosphorylated Mer2 at the S30 position. The primary antibody was used at a 1:5,000 dilution.
